Zelluläre Immunologie: Immunregulation und Zelltod im Immunsystem. Entstehung von humanen Autoimmunerkrankungen.

Allgemeines

In diesem Forschungsbereich untersuchen wir grundlegende Mechanismen, die zur Entwicklung von Autoimmunerkrankungen führen. Bei diesen Erkrankungen richtet sich das Abwehrsystem fälschlicherweise gegen körpereigene Strukturen.

Uns interessiert vor allem

  • die Rolle von Lipidmikrodomänen (winzigen Bereichen auf der Oberfläche von Proteinen) bei der Aktivierung von autoreaktiven (gegen körpereigene Strukturen gerichtete) T-Lymphozyten,
  • warum es zum Funktionsverlust von regulatorischen T-Zellen kommt, die unter physiologischen Bedingungen ein Überschießen der Immunreaktion verhindern,,
  • wie der programmierte Zelltod (Apoptose) reguliert wird und in welchem Ausmaß sich bei diesem Prozess Abbauprodukte anreichern, die wiederumautoreaktive T- und B-Zellen stimulieren können.

In unserem Labor charakterisieren wir Größe und Zusammensetzung von Lipidmikrodomänen in T-Lymphozyten. Ziel ist es, Anzahl und Funktion der regulatorischen Zellen bei Normalspendern und Patienten mit chronischen Autoimmunerkrankungen zu bestimmen. Regulatorische T-Zellen lassen sich im Labor vermehren, so dass es möglich ist,  ihre Membranrezeptoren zu untersuchen. Die Regulation des programmierten Zelltodes wird untersucht an Zellen von Patienten mit Autoimmunerkrankungen, vornehmlich Systemischem Lupus Erythematodes (SLE). Ein besonderes Merkmal dieser Krankheit ist die Entwicklung von Autoantikörpern gegen Histone und doppesträngige DNA. Wir wollen herausfinden, warum Abbauprodukte des Zellkerns, die während der Apoptose entstehen, eine Immunantwort auslösen Antigenen . Diese Untersuchungen sollen helfen, die Mechanismen besser zu verstehen, die zur Entstehung von Autoimmunerkrankungen führen und auf dieser Basis neue Therapieformen entwickeln.
