Arbeitsgruppe Endometrium & Implantation

Die Gebärmutterhöhle wird von einer ca. 1 cm dicken Gewebeschicht, dem Endometrium, ausgekleidet, die alle 4 Wochen im Rahmen der Menstruationsblutung abgestoßen wird. Bei Eintritt einer Schwangerschaft wächst der Embryo etwa 5 Tage nach dem Eisprung in das Endometrium ein und bildet dort den Mutterkuchen, die Plazenta. Diese Phase des Einwachsens, die auch Implantation genannt wird, ist äußerst sensibel und kann verschiedenen Störungen unterliegen, die zu einer Abstoßung des Embryos führen.

Da eine endometriale Implantationsstörung für einen unerfüllten Kinderwunsch und für die begrenzte Erfolgschance bei IVF- und ICSI-Behandlungen mit verantwortlich ist, untersuchen wir in unserer Arbeitsgruppe systematisch die Funktion des Endometriums und dessen Regulationsstörungen mit Hilfe von molekularbiologischen und proteinbiochemischen Untersuchungstechniken. Unsere Untersuchungen konzentrieren sich auf die verschiedenen Stufen der endometrialen Implantation (siehe rechte Spalte).

Stufen der endometrialen Implantation

  1. Anhand von Gewebeproben und im Zellkulturmodellen wird die Produktion und Regulation verschiedener Faktoren -wie z.B. pro-inflammatorischer Zytokine- untersucht, welche für die endometriale Vorbereitung der Implantation bedeutsam sind.
  2. Des Weiteren werden Adhäsionsproteine wie Integrine, Osteopontin, Galektine, Syndekane u.a. analysiert, welche die erste Phase der Implantation und somit die Anheftung des Embryos an das Endometrium regulieren.
  3. Die Wechselwirkung des Embryos mit dem mütterlichen Endometrium bei der fortschreitenden Implantation wird im Zellkulturmodell imitiert und analysiert.
  4. Bei Patientinnen mit Implantationsversagen oder wiederholten Fehlgeburten werden in endometrialen Biopsien verschiedene Funktionsmarker charakterisiert um endometriale Funktionsstörungen in der späten Implantationsphase zu analysieren.
  5. Im Zellkulturmodell wird der lokale Effekt von Medikamenten, z.B. Metformin, ein Antidiabetikum, welches in der Reproduktionsmedizin zum Einsatz kommt (z.B. bei PCOS), auf endometriale Zellen betrachtet.
